Default Emil Hansen - serial dilution techniques - RFI

Has anybody on the list a URL to a web resource which details the techniques
used by Hansen (or a modern equivalent) to isolate single cells of yeast.

Lest you think me an idle Brit I have spent much time Googling with the string
<"emil hansen" techniques yeast "serial dilution"> and many variations, adding
in "laboratory" "microbiology" and "beer".

Hi John;

Yes, I've diluted a culture for microscopy. I did it out of curiosity and
to see how well I could ID and sort out the critters involved--a 1% aqueous
solution of methylene blue sure helps...

What is it that you'd like to know?

Perhaps this s/b an off-line discussion...?

I think it's on topic. Feel free to discuss it in this group,
at your option of course.

I've had my nose in several microbiology books trying to find
the best way to do this myself, so I'd be interested.

looked at some yeast cells but I don't see how I could speciate them at
normal magnifications.

Ironically, John, the science of studying fungal and plant spores and
pollen etc., is palynology (correct spelling, even after a couple of glasses
of claret).

Google "streak-plate". You may find, for instance,
http://www.austin.cc.tx.us/microbugz/18streakiso.html

Another such technique is "pour-plate". That is consistent
with "serial dilution" but is more appropriate to counting
microorganisms than to isolating single colonies.

Those techniques are appropriate to microbiological labs,
but hardly to kitchens.

Bread and beer can be made with very little knowledge of=20
microbiology. But posters to this thread may be interested in

Try the n.g. sci.bio.food-science

or

sci.bio.microbiology


When I recall my home beer making, I seem to recall Hansen used a
"polarized" microscope. But I'm recalling this from memory of over 15
years ago.
